标题[求救] 胺基酸的衍生化

各位版友大家好：
我第一次做胺基酸衍生化，目标是将gama aminobutyric acid经PITC(phenyl
isothiocyanate)衍生化後以HPLC分析定量。但依照paper上的protocol作衍生化，一直得
不到预期的结果。
参照的步骤如下：
Standard derivatization
(减压离心浓缩去除溶剂的过程：37 °C)
˙50λ标准品(0.0125, 0.025, 0.05 mg/mL) 减压离心浓缩到透明
˙加入20λ coupling reagent (methanol:water:triethylamine 2:2:1)
˙混和後减压离心浓缩到透明
˙加入30λ PITC reagent (methanol:water:triethylamine:PITC 7:1:1:1)，
於室温下反应20分钟
˙减压离心浓缩去除PITC (eppendorf有白色固体残留)
˙PITC衍生物以500λ bufferA回溶
˙经0.2λ Millipore membrane过滤
HPLC analysis
˙column: C18 reversed-phase column (Nucleodur 100-5 C18 ec 5U, 250 x 4.6 mm)
˙operating temperature 43°C
˙UV absorption spectrum：254 nm
˙以gradient进行分析
Buffer A： 0.1 M ammonium acetate
Buffer B： 0.1 M ammonium acetate:acetonitrile:methanol (44:46:10, v/v/v)
BufferA、bufferB皆以acetic acid调整pH到6.5
时间 buffer A 比列
0-12.5 min 100-75%
12.5-25 min 75-50%
25-37.5 min 50-25%
37.5-50 min 25-0%
HPLC图谱中没有标准品的peak。
想请问是否有版友做过胺基酸的衍生化，
可以给一些建议，或是操作上需要注意的地方。
