标题[求救] 请问mitochondria isolation方面的问题

各位大大好~
我目前在做有关mitochondria相关的实验
目前的假设是 当加药之後mitochondria中的cytochrome c会被release出来
目前已使用kit进行完isolation的步骤 分出来的有分mitochondria以及cytosol两部分
请问在使用Western blot确认isolation是否成功的时候是在两条lane上loading同样的蛋
白量然後看tubulin(cytosol中有 mitochondria中没有) 和COX-4(cytosol中没有 mito-
chondria中有)这样子吗? 还是只看COX-4就可以了?
因为目前我所遇到的问题是:
在tubulin结果中看到isolation不成功(虽cytosol中较多 但跟mitochondria中差距不大)
但在COX-4结果中看到isolation成功(mitochodria中较多 且cytosol中几乎没有)
请问我该相信哪一个呢?
