標題[求救] Hot marker & Sequencing gel

請教大家
小弟目前正在製作有P32 label的DNA marker
使用的是 ΦX174 /Hinf I 的marker, 用PNK進行5'end-label
之後用CentriSpin 20 進行purification,去除掉沒label上的ATP
然後run 8% sequencing gel(含7M urea)
可是不知道為何小片段的marker(24bp, 48bp, 60bp)看起來總是很模糊
是因為我在pruification的過程中lost小片段marker?
還是我應該用fill-in的方式來label?
還是gel本身的問題?
