※ 引述《sirius1970 (喪失.........)》之銘言： : 看了幾篇 不過還是抓不到老師要的重點 : 有沒有人知道老師要的東西到底是什麼 : 是轉一個gene進入署germ line內 然後看他署子代對此DNA methylation的變化嗎 : 還是做 somatic nuclear transfer然後看子代變化 : 請大家就救急囉 在進行animal cloning時 由於利用的核 是somatic cell 在這樣的細胞核中 [很多基因已經被methylation]而失去功能 ex. 在epidermal cell中 不會表現gland cell的基因 而胚胎發育時 必須使每個細胞回覆到totipotent的狀態 亦即使每個基因都有表現的能力 而在細胞分化時 才可藉有正確的 re-methyltion重新定義每個細胞執行的功能 但是在cloned animal offsprings上 往往無法去除 nucleus donor原本的methylation ex. 利用fibroblast cell當作nucleus donor結果 cloned cytoblast仍呈現 fibroblast的methylation pattern. 這樣使得有些基因由於延襲了donor 的methylation pattern而無法正常表現 故這樣的cloned cytoblast沒有正確發育的能力 可以用 abnormal methylation + cloned animal關鍵字 --
