※ 引述《sirius1970 (丧失.........)》之铭言： : 看了几篇 不过还是抓不到老师要的重点 : 有没有人知道老师要的东西到底是什麽 : 是转一个gene进入署germ line内 然後看他署子代对此DNA methylation的变化吗 : 还是做 somatic nuclear transfer然後看子代变化 : 请大家就救急罗 在进行animal cloning时 由於利用的核 是somatic cell 在这样的细胞核中 [很多基因已经被methylation]而失去功能 ex. 在epidermal cell中 不会表现gland cell的基因 而胚胎发育时 必须使每个细胞回覆到totipotent的状态 亦即使每个基因都有表现的能力 而在细胞分化时 才可藉有正确的 re-methyltion重新定义每个细胞执行的功能 但是在cloned animal offsprings上 往往无法去除 nucleus donor原本的methylation ex. 利用fibroblast cell当作nucleus donor结果 cloned cytoblast仍呈现 fibroblast的methylation pattern. 这样使得有些基因由於延袭了donor 的methylation pattern而无法正常表现 故这样的cloned cytoblast没有正确发育的能力 可以用 abnormal methylation + cloned animal关键字 --
