Guidelines and assignments for Mol Biol class on Dec 14, 2010 Reading: Single molecule-sensitive probes for imaging RNA in live cells. ( http://imm.med.ncku.edu.tw/news.htm ) 公告事项中 PBL课程资源两个PDF档为此篇paper及supplementary! 1.Grouping: IMM and MT students are divided into 3 groups each (i.e., IMM1,IMM2, IMM3, MT1, MT2, and MT3) 2.Prepare ppt slides (the whole group, each slides with names of th contributors) for no more than 25 min presentation time, turn in by e-mail to both Dr. Wang ([email protected]) in IMM and Dr. Young ([email protected]) in MT no later than 4 pm on Monday (Dec. 13) 3.Problem based learning for each group: Each group needs to search more information from the PUBMED to clarify the assigned problems/topics in the list below. Please simplify complicated background with cartoons and some text explanation If possible, you may use the following outlines to describe the figures of the paper in details: 1. Background and Aim 2. Experimental strategy 3. How to prepare the experimental materials 4. Instruments (optional: available in NCKU ?) and methods 5. Results 6. Conclusion and impact IMM1 Q1. Describe the key concepts involved in MTRIP methodologies and the optimization strategies (Fig 1. a-c). Compare the difference between MTRIPs and MS2-GFP binding systems in detection of intracellular RNA molecules. MT1 Q2. Please describe MTRIP sensitivity with experimental model on glass surfaces and in the cytosol. Please interpret the results in Fig 1. d, e and suppl Fig.1. IMM2 Q3. Please describe the types (or composition) and functions of “cytoplasmic RNA granules” and how these mRNA-protein organizations (mRNPs) regulate mRNA. MT2 Q4. How did the authors use MTRIP to visualize and quantify beta-actin mRNA in A549 cells ? Please describe the instrument of time-lapse microscopy. How did the authors record the tracking of beta-actin mRNA in A549 cells? (Fig.a-d,suppl video 1-3) ? IMM3 Q5. How did the authors use MTRIP to demonstrate (1) the co-localization of beta-actin mRNA and its binding proteins (suppl Fig. 2-4); and (2) the specifi detection of hRSV genome (suppl Fig. 5-7) . Please explain in details how to correlate the imaged molecules with their intensity and distribution (you may need to introduce the Manders overlap coefficient and the Wilcoxon-Mann-Whitney test.) MT3 Q6. How did the authors use MTRIP for the study of RNA-protein colocalizaiton in live cells by using RSV RNA and TIA-1 in the absence and presence of stress. (Fig. 3a-d, suppl Fig 8, 9, suppl video 4-8). --
