标题[求救] 关於permanganate footprinting

敝实验室最近尝试做permanganate footprinting
就是利用KMnO4会氧化T使其之後能被piperidine切断的特性来侦测protein binding site
一般的footprinting都会接上istope-labeled primer进行LM-PCR
而小弟我想先用一般primer先试试看有没有办法有效P出东西
但不管怎样做都只得到一堆smear的杂band却无法在想要的位置看到很亮的PCR product
想请问一下是依定要用isotope-labeled的primer才看的到东西吗?
或者是其他原因导致我无法得到产物?
