标题[求救] 从活体器官纯化GST

取的器官是肝
均质化後溶於phosphate buffer (25ml, 10mM, pH7.0)
Sonication on ice for 2min ,then centrifuged at 4ﾟC and 9000 rpm for 25min.
Supernatant 经过Filter後
用glutathione-Sepharose 4B binding 30min
用phosphate buffer (10ml, 10mM, pH7.0) 洗
接着用phosphate buffer(30ml, 20mM, pH7.0), containing NaCl (150mM)
最後用Tris-Cl buffer (5ml, 50mM, pH8.0), containing GSH(10mM) 去做Elution
的动作
最後在用Gel-filtration (S-100 column
但是最後跑胶後 在接近的位置有两条band
目前判断是有两种protein (其一应该是GST
不晓得这样的步骤是否有问题@@
因为主要是想但纯得到GST
