标题[问题] shRNA之transient transfection及stable transfection

目前使用的shRNA是购於中研院
有表示其knockdown效率高达96%
但我用shRNA做transient transfection
使用lipo2000 RNAi transient tranfection 到HNK293 cell
隔天换新鲜medium培养，三天後取细胞total RNA
最後做RT-PCR 结果knockdown效率超差
所以想请问板上是否有人做过类似实验?
在transient transfection之前，shRNA是否有何处理?
或使用哪种liposome? transfection时细胞几成满?
等等详细流程，先在此感谢各位回覆
