标题[求救] 紧急求救! 实验结果(+)和(-)值过於相近却找不出原因

昨天我作了转录因子AP-1的分析，最後判读的机器采用冷光分析仪。
按照protocol的结果，positive reagent control组应为
844250, 853300,783750(RLUs)，
得Mean值为827100。
negative reagent control组为
9166, 9162, 9817(RLUs)
得Mean值为9382。
但不幸的是跑出的data结果：
positive reagent control 平均值为9万多
negative reagent control 平均值为4万多
(老板的脸都绿了....)
我的推论是：1.不同的组织细胞跑出的数据也不同？
2.protocol没有提到要用多少波长，我参照文献选择625的波长，
不同的波长有可能会影响结果的判读？
3.实验过程中可能污染?
但以上推论老板都不接受，请教各位专业大大，我该如何或是用什麽方式
解释这样的结果呢??重作大概也来不及了....请各位帮帮忙!!
