标题[问题] 关於 yeast two-hybrid vector

最近在接 yeast two-hybrid 用plasmid
用的vector是pGBKT7 7.3kb

接一个1.2kb的insert 切位 pst1 bamH1
PCR後单切 胶回收 再单切 再胶回收
接的时候先45度C Denature Vector insert
再跟据跑胶ban的亮度去决定比例
total 20 ul
然後10~16度C 水浴 O/N
隔日transform到 DH5a 的E.coli中表现
讲了这麽多....反正就是接不进去
学长说可能是因为vector很长所以两端接不太到~~
有克服的方法吗??
