有几个跟Cy5有关的问题 希望知道的人能够给点指点^^ Red laser light that produces fluorescent emission from Cy5 can also switch the dye to a stable dark state. Exposure to green laser light converts Cy5 back to the fluorescent state, but the recovery rate depends critically on the close proximity of a secondary dye, Cy3. 主要是有关这一段的问题.在看到这篇paper以前我认知中的Cy5是照红光就会亮, 然後就一直亮到它photobleach为止.所以我看不懂为什麽red laser会 "switch the dye to a stable dark state".而且这里green laser的存在目的是什麽呢? 如果不照green laser, 一直照red laser, Cy5就不会再亮了吗? "green laser light converts Cy5 back to the fluorescent state", fluorescent state不是指excited state吗?可是在paper里面照绿光的 时候是没有emission的. 这几部分我真的很疑惑. 另外请问Cy3跟Cy5之间的距离会影响recovery state的问题,这应该从哪个角度去解释呢? 因为我查相关资料,里面也只是提到这个现象,但并没有详细解释为什麽? 拜托大家了^^ --
