stem cell, 又称为干细胞, 其定义为只要具有differentiation以及 self-renew的能力的细胞即可称之为stem cell.differentiation比较简单 就是细胞由一种型态转变成另一种phenotype, 其中可能牵涉到一些新的蛋白 及基因表现。stem cell的细胞分裂有两种, symmetry cell division以及 asymmetry cell division, symmetry cell division可以增加stem cell的数量 而asymmetry cell division就是一边产生differentiated cell而另一边 产生一个活性跟原来的干细胞活性相当的干细胞, 这就是"self-renew"。 而在干细胞执行differentiation及self-renew时，周边的microenvironment非常重要 就是所谓的"niche"，其可以提供stem cell执行功能时所需要的养分， 如neural stem cell其周边的endothelial cell即负责这个角色! 就stem cell的function来说, 又可分为三种, pluripotent stem cell， totipotent stem cell，multipotent stem cell，在受精卵开始发育後大概 4天时,会成为一个8细胞体，此时这8个细胞每一个细胞都是totipotent stem cell ，其功能最为强大，每个细胞都能发展成一个完整的生命体!若发育到行成 blastocyst时，内层细胞团包含着pluripotent stem cell，此时每个细胞都 可以发育成生命体的任何一种器官!最後一种呢，multipotent stem cell-- 就是成人干细胞，这就不需要我多说了吧...mesenchymal stem cell， neural stem cell，HSC等等都是论文常提到的。 还有一个比较特别的 也是最近渐渐被注意的，就是Cancerous stem cell 我之前看的一篇PANS说到，Cancer cell为什麽经过放射腺及药物治疗後 过几天又开始活起来了??!!那篇paper提到一个论点就是cancer cell存在着 cancerous stem cell，因为此种stem cell跟一班的stem cell一样，会表现 ABC transporter，熟的人都知道这是一种drug-resistant transporter，可以将 药物往细胞外送，减少对细胞的伤害，而那篇PNAS的论文也将Cancer里的 cancer stem cell进而纯化出来，发现他的确会造成mestasis，而普通cancer cell并不会。现在非常多的paper尤其是nature系列的常提到这种干细胞! 喔喔 今天先打这样吧 过几天来谈谈 stem cell therapy 那以下是我seminar要讲的paper: Hematopoietic Stem Cells Convertinto Liver Cells within Days without Fusion NATURE CELL BIOLOGY VOLUME 6 | NUMBER 6 | JUNE 2004 请多多指教搂 --
