IMPACT OF NORMAL AND PATHOLOGICAL ACTIVITIES ON KCC2 MOBILITY AND FUNCTION IN HIPPOCAMPAL SLICES The K+/Cl? cotransporter KCC2, which extrudes intracellular chloride controls chloride homeostasis that critically determines the polarity and efficacy of GABAergic transmission in the brain. Our focus is to understand the mechanisms that control KCC2 membrane expression and function in order to understand ?and possibly interact with? the functional consequences of a brain trauma or epileptic activity in the adult brain. The proposed project is to study the membrane dynamics of the KCC2 transporter in integrated network of cortical neurons, and to examine how these properties can be modulated under normal and pathological epileptic conditions. This project will require developing the Single Particle Tracking technique in brain slices. For this purpose, the post?doctoral candidate will collaborate with biochemists and specialists in biomedical imaging to develop Single Particle Tracking technique in an integrated neuronal network. The lab is located in the heart of Paris in a rich neuro?scientific environment. The IFM houses state of the art research facilities (bi?photon microscope, several confocal microscopes for fixed preparation and live cell imaging, electrophysiology rigs, single particle tracking rigs coupled with electrophysiology, ratiometric fluorescence and photolysis system as well as molecular biology and imaging platforms). Preference will be given to candidates with a track record of peer?reviewed publications, extensive experience with cell imaging.
