3. 「实验微免」虽然是下学期的课~却要分一学年的时间去跑各各实验室来完成~ 　　由於这没有电子档给大家~在这里羊羊先简单列一下项目~大家可以回去问问学长姐看需要修什麽？ 刘校生老师：Transient and stable transfection 选择本实验者需於无菌操作下，将beta-galactoseidase报导基因 ，已转殖的方式(calcium phosphate)送入293细胞中，并有1%以上之基因 表达者时，才算完成此实验认证。 需先完成(4)和(8)之训练方可进行此实验。 黎焕耀老师：Flow cytometry 选择流式细胞仪技术课者，须先接受必帝公司流式细胞仪一天的 讲习课程之後，於黎焕耀老师实验室安排的时间，根据下列的实验流程 ，亲自做小鼠脾脏细胞或人周边血液，分析T-淋巴球及B-淋巴球细胞的 比例，其结果在范围之内，才算完成此技术的认证。 林以行老师：Confocal microscopy 　　　　　　(需修课费用1500元，可以先询问指导教授是否愿意补助) 选择共轭聚焦显微镜技术课程者，必须先参加由医技系黄辉升老师主持 之成大医学院核心实验室之共轭聚焦显微镜使用说明会，并接受实机操 作观摩(共约2小时，另加自行练习时间）。参加实机认证考试前，可选 由本实验室成员 (已通过认证者包括一名super user及四名individual user)之协助下进行实机指导以便熟悉实验操作後，可直接由super user 进行技术认证，包括染色方法、共轭焦显微镜操作、标的影像撷取与分析 等技术，认证合格者除成为individual user之权限，亦完成本部分课 程之学分取得。) 何涟漪老师：Gene cloning 选择本课程者，须将beta-gal 基因以PCR增辐後，插入质体载体，将重组 质体送入大肠杆菌，得到转型成功的菌株，并抽取其中重组质体，检查其 限制酶切割图谱无误，方得此技术的认证。 萧瑷莉老师：PCR & RT-PCR 选择PCR及RT-PCR技术课程者，将实验室提供cDNA模板，设计两端引子并 依据实验流程放大增幅此一片段。此外抽取组织细胞样本RNA，利用 reverse transcriptase将其反转录为cDNA，再利用PCR增幅此一片段。 实验结果其产物讯号强度及片段大小必须符合要求，即可完成此技术认证。 余俊强老师：Animal experimentation Tissue preparation for general morphology examination and immunohistochemical staining 学员需先观看动物实验技术教学光碟及阅读相关操作资料了解基本观念， 然後由本实验室成员示范指导，以活体小鼠进行练习，熟悉包括尾静脉 注射、腹腔注射、皮下注射、肌肉注射、眼眶采血、心脏采血、灌流、 脏器采摘、固定及包埋等技术。认证方式为亲手操作，并获指导人员认可。 陈舜华老师：Cell culture & Plaque assay 1. Cell culture：Students should be able to maintain and subculture a cell line without contamination for two weeks 2. Plaque assay：Students should be able to determine viral titers in samples by plaque assay on appropriate cells 凌　斌老师：Reporter assay Innate Immune Recognition of Virus Infection (Experimental Immunology in Pin Ling Lab) 1. Learn 293 cell culture and transfection 2. Learn viral infection or PRR stimulation by synthetic ligands 3. Learn the luciferase reporter assay 王淑莺老师：Protein expression and purification Students will learn how to express recombinant proteins in E. coli system and how to purify proteins using immobilized metal affinity chromatography (IMAC). Students should be able to obtain protein with high purity. --
