protein (cold and heat denaturation). Plot the temperature-dependence of ΔH

2.Consider a two-state system with energy levels, E1 and E2. The system has 1
mole distributed between these two energy levels, E1 and E2.

rement of Trp fluorescence intensity (arbitrary unit) as function of tempera-
ture for a pair of wild-type (WT) and mutant Pfu DNA polymerase. This basica-
lly is a protein melting curve we dealt with in class.
(1a). What are the melting temperatures of WT and Mutant proteins, roughly?
You may assume protein at 10°C is totally folded and protein at 95°C
is totally unfold.
(1b). Explain how you will use the appropriate plots and steps needed to det-
ermine ΔH, ΔS of unfolding these two proteins (no calculation requir-
ed).
(1c). Assume ΔH of the WT is 23.8 Kcal/mol, and ΔS is 78 cal/K-mol. If you
