3.In the lecture of Botanical Electron Microscopy, we have discussed a lot
about three dimensional images of macromolecule and tissue. Could you
correlate their instrumentation, method and application.
The three dimensional appearance of SEM images is due to differences in
contrast between various structural feature of the specimen when they are
displayed on the viewing monitor. And the three dimensionality and contrast
are due to the yield of secondary electrons from various parts of the
specimen.
Stereo imaging with the SEM
Application:
1. The enhancement assists the viewer in discriminating between projections
and depressions, and , it elucidates spatial relationship to help determine
distances between two objects.
Method: (generate stereo pairs and merge the separate views )
1. Generate two different micrographs from separate viewpoint
Generate stereo pairs
a. shifting the specimen slightly to the left or right
b. rotating the specimen
c. deflecting the electron beam electronically
d. tilting the electron beam electronically-------most prevalent
example:(using tilt to generate stereo view in the SEM)
1.Take the first micrograph in the usual manner.
2.With out moving the specimen and trace the out line of the specimen on
the CRT
3.Tilt the specimen slightly.
4.Refocus the specimen using the Z control adjustment of the stage.
5.Realign the image.
6.Adjust the brightness and contrast of the image in the SEM to match the
first micrograph and take the second micrograph.
2. Merge these separate views using the appropriate tools
(1)Print the two separate images at the same contrast and density levels in
the dark room.
(2)The micrographs are then placed alongside each other and viewed with a
special set of glasses(stereo viewers) placed directly over the two prints.
我又来讲解了
这个题目主要是要问说什麽是three dimensional image?
而我要如何去制作这个影像(method)
而他有什麽功能(application)
所用到的仪器(instrument)
我们主要是利用SEM image所产生的contrast,结合两张影像利用人类的视差
(parallax phenomenon)使得本来是2D的照片看起来却向是有3D的效果
而这主要的优点就是可以让我们清楚的了解我们看到的影像中哪些是凸的哪些是凹的
并可以让我们知道样本之间的距离。
而如何去取得stereo pairs就是将specimen 稍微tilt一下取得两张影像
在利用我们一般看立体图的方法去观察这样就可以取得我们想要的3D立体图了
或者是利用stereo viewer的帮忙。
