1. Multi-hit model
In the multi-hit model, cancer arises by a process of clonal selection.
As mutations accumulate in any progeny, the cell gains the ability to
grow and divide uncontrollably and to metastasize.
Decades are required for these multiple mutations to occur
2. Two-hit hypothesis:
According to Knudson's two-hit hypothesis, the development of tumors,
such as retinoblastoma, requires two separate mutations. In hereditary
retinoblastoma, the first mutation or first hit is inherited, and the
second is acquired somatically.
